Cowboys tt hasn't had much in the way of dt's, so I've tried a couple of different things. 

Today I put my 83 d-law next to merlin Olsen in nickel and wow! Somehow they are working together to produce pressure that I couldn't get before with 91 mean joe! 

There doesn't seem to be a negative effect to my tt Chem, and I don't see anything ex'd out. So I'm rolling with it. 

My question is why does other oop setups sometimes penalize me on Chem? How do I know what works and what doesn't on oop?

It is probably because you made the change in the specialists tab.  The specialist tab do not count towards Theme teams chems.   By putting cowboys Dlaw in at rush DT1 he will not be penalized But if you are in the Defense tab and put him in at DT1 instead of LE then you will get "Ex'D" as you say.  
-Sidenote:If you get the 89 DLaw at any point you can power him up and he has secondary of RE so you can put him either RE slots and not lose the Cowboys chem, if you have 2 other cowboy LEs.
-Also if you manually sub him into DT when the game start or use some kind of rush/speed package you can get Dlaw into DT without TT penalties as well.

Putting someone oop anymore does nothing except make the chem not work on the first 2 tabs (which usually isnt a big deal, if youre trying to hit 40 or 50/50 then maybe). If they have the ability to go to another spot via chem you can pay 500 training (which is expensive right now) and keep the chem. 

If you go to the specialist tab, you can put anyone in no penalty or if you sub them in during the game theres no penalty either.

if they dont have a secondary position where you put them it’ll disable the chem for that player

You still have to buy that chem to put them there too. But if its like CB and moves to safety and you have safety strat card they wont get boosts even if you play them there and have the chem unlocked.
