Manual I'm pretty sure as it appears you can only double one player at a time
Try a solo challenge. Equip an oline player with post up only and see if it lights up during a double team you didnt commit. Pretty sure ive seen it light up without committing but I cant test atm.

As with many things, it depends. If there's just a three-man pass rush, then your O-line will typically double team automatically. So yes, anytime your blocker is engaged in a Double Team, that Post Up ability will just work. It's best if you manually set your blocking adjustments, but that's not something that necessarily needs to happen on each and every single down.
Post Up on the Center is fantastic, however, the ideal scenario is to have Post Up on both guards, which will ensure that no matter what way the double-teams happen, they will always become dominant.
